An enzyme that follows Michaelis–Menten kinetics gave these initial rates (v0): 20, 33, 50, 67, 80 µM/s at substrate concentrations of 0.5, 1, 2, 4, 8 mM. Which value is closest to the enzyme’s Km?
Pick one. The red pen does the rest.
Why each choice fails
- ASays more than the data: 1.4 mM is roughly where v0 would reach 40, half of the highest measured rate. But v0 is still rising at 8 mM, so 80 is not Vmax.
- BHolds: at [S] = Km the rate is half of Vmax.
- CWrong variable: 4 mM is half of the highest [S] tested. Km is the [S] at which the rate, not the substrate concentration, is half of its maximum.
- DLook-alike terms: 8 mM is the [S] nearest saturation, which bears on Vmax, not Km. There the rate is already 80% of Vmax.
